The Alzheimer's Disease (AD)-Detect® Beta-Amyloid 42/40 Ratio, measured in plasma, is a diagnostic tool used to assess the ratio of beta-amyloid proteins 42 and 40. Abnormal levels of this ratio may indicate an increased risk of Alzheimer's disease.

Beta-amyloid proteins are peptides that are produced throughout life, with a crucial role in the development of Alzheimer's disease. Beta-amyloid 42 (Aβ42) and beta-amyloid 40 (Aβ40) are two variants of these proteins.

Aβ42 is particularly significant because it is more prone to form aggregates and plaques in the brain, which are hallmarks of Alzheimer's disease. In contrast, Aβ40 is more soluble and less likely to form these aggregates.

The ratio of Aβ42 to Aβ40 in the bloodstream is indicative of the balance between these two forms. In Alzheimer's disease, this ratio tends to be altered, with higher levels of Aβ42 relative to Aβ40. This imbalance is thought to contribute to the pathology of the disease by promoting the aggregation of beta-amyloid peptides in the brain.
